Doxophylline is a medication that is used to treat asthma, COPD, and bronchospasm.
Doxophylline is an experimental bronchodilator. The inclusion of a dioxolane group at position 7 distinguishes it structurally from Theophylline. Doxophylline inhibits phosphodiesterase 4 specifically, relaxing bronchial smooth muscle. However, unlike Theophylline, Doxophylline appears to have lower affinity for adenosine A1 and A2 receptors, which may explain for the drug's improved safety profile. Doxophylline has been shown to suppress platelet activating factor (PAF) and leukotriene synthesis.
Elderly: 200 mg tablet two or three times daily.
Adults: 400 mg tablet two or three times daily or as prescribed by the physician.
Children:
If required daily dose of Doxophylline is 400 mg then Doxophylline SR tablet to be taken once daily or as prescribed by the physician
Doxophylline should not be used with other xanthine derivatives. Xanthines have been shown to have toxic synergism with ephedrine. Concomitant therapy with troleandomycin, lincomycin, clindamycin, allopurinol, cimetidine, ranitidine, propranolol, and anti-flu vaccination, like other xanthines, may reduce hepatic clearance of xanthines, resulting in an increase in blood levels. There is no evidence of a link between Doxophylline serum concentrations and harmful occurrences.
Doxophylline is not recommended in cases of acute myocardial infarction. It is also not recommended for individuals with hypotension, nursing women, or those who have demonstrated hypersensitivity to its components.
Doxophylline seldom produces significant adverse effects, although the risks are comparable to those associated with excessive coffee use. Nausea, vomiting, headache, upset stomach, and heartburn are some of the symptoms.
Doxophylline does not cause fetal damage when fed to pregnant animals, and it has no effect on reproductive ability, according to animal reproduction studies. However, because human experience with xanthines during pregnancy is limited, pregnant women should only be administered xanthines if absolutely necessary. Doxophylline is not recommended for breastfeeding moms.
A number of known factors impact the half-life of xanthine derivatives. Patients with liver illness, congestive heart failure, and those on certain other treatments such as erythromycin, troleandomycin, lincomycin, allopurinol, cimetidine, propanolol, and anti-flu vaccine may have it extended. In these instances, a reduced Doxophylline dosage may be required. Phenytoin, other anticonvulsants, and smoking may accelerate clearance, resulting in a shorter mean half-life. Doxophylline at greater dosages may be required in certain instances.
