Glucosamine Sulfate + Diacerein
This tablet is indicated in-
Glucosamine: Glucosamine (2amino2deoxyalphaDglucose) is a naturally occurring amino sugar, produced by the body and found in some foods. It is the most basic building block required for the biosynthesis of glycosaminoglycans (GAGs) such as hyaluronic acid, keratan sulfate, and chondroitin sulfate. GAGs bind to proteins and form proteoglycans, an essential structural component of articular cartilage. When the cartilage in the joints degrades, the process of joint degeneration develops. It also helps in the formation of ligaments, tendons, nails and many other connective tissues. When we supplement with artificially synthesized glucosamine sulfate, it increases the glucosamine levels in the body, thereby facilitating cartilage production and repair. Glucosamine also activates chondrocytes to help produce GAGs and proteoglycans.
In humans, approximately 90% of glucosamine, used as glucosamine sulfate, is absorbed from the gastrointestinal tract. Metabolized mainly by the liver and excreted in the urine.
Diacerein: Used to treat osteoarthritis. It also has analgesic, antipyretic and anti-inflammatory activity. It releases in vitro and directly inhibits the synthesis of InterLeukin1 (IL1), which is the main cytokine involved in cartilage destruction. Because of its specific mode of action, it has been shown to have disease-modifying effects in experimental models of osteoarthritis and in humans with osteoarthritis of the fingers and knees.
Oral bioavailability of diacerein 56%. Food intake also slowed the time to peak concentration but was associated with a 25% increase in absorption. Therefore, diacerein is best taken with food. Mainly related to the protein albumin. Diacerein is extensively (100%) metabolized in the liver after oral administration. Urinary excretion of diacerein in the form of its metabolites varies from 35% to 60%.
Use in adults: One tablet twice daily with food.
Use in children and adolescents: The safety and effectiveness of children and adolescents under the age of 18 years have not been established.
Decrease absorption of Diacerein with aluminium and/ or magnesium hydroxide antacids. Increase risk of diarrhea with laxatives or antibiotics.
Contraindicated for those who shows hypersensitivity to Glucosamine or Diacerein.
No serious adverse effect has been reported. Nausea, vomiting, diarrhea, epigastria pain, headache, skin rashes & intense yellow coloring of urine may be occurred.
This tablet is contraindicated during pregnancy and breastfeeding.
Caution should be practised when administering this tablet in case of patients who are allergic to Glucosamine or Diacerein.
