Dyspeptic symptom complex, frequently accompanied by delayed stomach emptying, gastroesophageal reflux, and esophagitis:
Acute nausea and vomiting of functional, organic, infectious, or dietary origin, or caused by radiation or medication therapy, or caused by migraine. Parkinson's disease is characterized by dopamine agonist-induced nausea and vomiting. Radiological investigations: Accelerating barium transit in subsequent radiological examinations.
Domperidone is a dopamine antagonist that works primarily by blocking dopamine receptors in the chemoreceptor trigger Zone (CTZ) in the stomach. Its gastroprokinetic activity is based on its ability to inhibit dopamine receptors, which regulate gastrointestinal motility. Domperidone has practically little action on dopaminergic receptors in the brain due to its poor penetration across the blood-brain barrier, omitting psychotropic and neurologic adverse effects. Domperidone restores normal upper gastrointestinal motility and tone, aids stomach emptying, improves antral and duodenal peristalsis, and controls pylorus contraction. Domperidone also enhances esophageal peristalsis and decreases esophageal sphincter pressure, preventing gastric content regurgitation.
Domin 10 mg tablet should be taken 15-30 minutes before meals and, if necessary, before retiring.
The usual recommended oral dose of Domperidone is as follows:
In dyspeptic symptom:
In acute and sub-acute conditions (mainly in acute nausea and vomiting):
By rectum in suppositories:
Domin 10 mg may lower the danger of bromocriptine's hypoprolactemic impact. Domperidone's effect on Gl function may be inhibited by antimuscarinics and opioid analgesics. Domperidone should be used with caution when combined with MAO (monoamine oxidase) inhibitors.
Domin 10 mg is contraindicated in individuals who have a history of hypersensitivity to the medication, as well as in newborns. Domin 10 mg should not be taken if there is a risk of gastrointestinal stimulation, such as gastrointestinal bleeding, mechanical blockage, or perforation. It is also not recommended for people who have a prolactin-releasing pituitary tumor (prolactinoma).
Domin 10 mg has the potential to cause hyperprolactinemia (1.3 percent ). This can cause galactorrhea, breast enlargement, discomfort, and a decrease in libido. During domperidone therapy, dry mouth (1%), thirst, headache (1.2%), anxiety, sleepiness (0.4%), diarrhea (0.2%), skin rash and itching (0.1%) are possible side effects. Clinical investigations show that extrapyramidal responses occur in 0.05 percent of individuals.
Domin 10 mg tablet's safety has not been established, and it is thus not recommended during pregnancy. Teratogenicity in the fetus has not been shown in animal investigations. Domperidone can cause galactorrhea and enhance postpartum lactation. It is released in breast milk, but in such tiny amounts that it is not considered hazardous.
Domin 10 mg should be used with extreme caution in children due to an increased risk of extrapyramidal effects in young children due to an inadequately formed blood-brain barrier. Domperidone, because it is extensively metabolized in the liver, should be taken with care in patients with hepatic impairment.
